چکیده

The distribution and renewal of RNA in mature rod visual cells has been studied by histochemical and autoradiographic technique in the rat and the frog. Staining with methyl green and pyronin, combined with ribonuclease extraction, revealed the presence of RNA in the nucleus and inner segments of rods in both species. Synthesis of RNA was studied by injecting cytidine-H, a radioactive precursor of RNA. Within 15 minutes after the injection, radioactive RNA could be detected in the nuclei of retinal rods in both the rat and the frog. RNA formation was not observed in other parts of the cell. By 1 hour after the injection, the new RNA began to move out of the nucleus and into the inner segment. This process was similar in both species, although it occurred more rapidly in the rat. These results demonstrate that RNA is continually renewed in mature visual cells and that inner segment RNA and nuclear RNA are both produced in the nucleus. The effect of prolonged bright light or total darkness on the renewal of RNA in retinal rods was studied in the frog. In both of these experimental situations RNA formation was depressed by a significant and similar amount.

Morphology of retinal photoreceptor layer in continuous light-exposed and dark-adapted male cats

The morphology of retinal photoreceptor layer was studied in continuous light-exposed and dark-adapteddomestic male cats (Felis catus). The eyes of 12 healthy adult cats (4 in continuous light-exposed group, 4 in continuous dark-adapted group, and 4 in control group) were routinely fixed and studied by electron microscope. Results showed that the general structure of photoreceptor layer in this...

The Renewal of Protein in Retinal Rods and Cones

The renewal of protein in retinal rods and cones has been analyzed by quantitative electron microscope radioautography in adult frogs injected with a mixture of radioactive amino acids. Protein synthesis occurs predominantly in the ergastoplasm, localized in the myoid region of the photoreceptor cells. Much of the newly formed protein next flows through the Golgi complex. In rods, a large propo...

Structure and arrangement of photoreceptors in the retina of big eye kilka, Clupeonella grimmi (Kessler 1877)

The big eye kilka, Clupeonella grimmi, is a marine fish living in depth of 20 to 200 meter of the Caspian Sea. Its eye and retina were processed for histological and SEM studies. Paraffin embedded retina was cut radially and tangentially in 5 ?m thickness and stained with hematoxylin and eosin method. The unstained sections were manipulated for SEM image observations. Tangential retinal sect...

Pluripotent stem cells as the cells with a capacity for self-renewal and differentiation into various specificcell types have been highly regarded in regenerative medicine studies. To repair the eye disease damages, thedifferentiation into retinal pigment epithelial cells of pluripotent stem cells has gained great importance inrecent decades because the inappropriate function of these cells is ...

Chloride channel protein 2 prevents glutamate-induced apoptosis in retinal ganglion cells

Objective(s): The purpose of this study was to investigate the role of chloride channel protein 2 (ClC-2) in glutamate-induced apoptosis in the retinal ganglion cell line (RGC-5). Materials and Methods: RGC-5 cells were treated with 1 mM glutamate for 24 hr. The expression of ClC-2, Bax, and Bcl-2 was detected by western blot analysis. Cell survival and apoptosis were measured by 3-(4,5-dimeth...
